A conversation with Greg, founder of Hestia Retreat, on the neurochemistry of total Darkness - and how the brain's natural pharmacy moves from melatonin to pinoline to DMT the longer the lights stay off.

We chatted with Greg - this time to go deeper into the neurochemistry. What exactly is happening inside the skull when the lights go out for days at a time, and what does the brain's own pharmacy have to do with ancient ideas of enlightenment?
The major cortical centres essentially get starved of their usual fuel. Robbed of stimulation, mental and cognitive function in the higher brain begins to quieten. But it's not a simple shutdown - emotional and feeling states become accentuated, the sense of smell sharpens, and perceptions that are normally dormant start to come online. Dreams become more lucid, and the dream state starts manifesting inside conscious awareness itself. Eventually we awaken to an inner sense of Source, of spirit, of soul. Taoists call this descent into the void. It's the Darkness of deep, inner space - and it's quite different to simply closing your eyes.
Yes - what Taoists call the 'Crystal Palace'. It's the collection of endocrine glands sitting at the very core of the brain: the pineal, the hypothalamus, the thalamus, and the pituitary. Mantak Chia, who's written extensively on Darkness technology, describes the pineal gland as initiating a cascade of inhibitory reactions that permit visions and dream-states to emerge into our conscious awareness. Eventually the brain synthesises what he calls the 'spirit molecules' - 5-MeO-DMT and DMT - facilitating what he describes as transcendental experiences of universal love and compassion. I should say upfront that hard science explicitly mapping this neurochemistry through a full Darkness journey is expensive and difficult to do. What we work with is a synthesis of educated theory, spiritual wisdom, and direct experiential reports.
In the absence of light, tryptophan evolves through a sequence of indoleamines in the Crystal Palace over time. The timeline varies between individuals and the shifts aren't sudden, but the general progression looks something like this: days one to three, melatonin. Days four to six, pinoline. Days seven to nine, 5-MeO-DMT. From day ten onwards, N,N-DMT. Tryptophan is the precursor for both serotonin and melatonin - serotonin speeds up mental activity for waking life, melatonin slows it down for the regenerative work of sleep. Without light, the Crystal Palace synthesises melatonin and triggers a cascade into the visionary tryptamines beyond it. Knowing that progression is genuinely useful when someone's deciding how long to stay in.
Melatonin regulates your circadian rhythms and sleep-wake cycle. It promotes sleepiness, helps synchronise your internal clock, carries antioxidant properties, and influences seasonal behavioural changes. Levels rise naturally in Darkness and fall in light - that's the basic mechanism. I think of this as the brain's first gift in Darkness: a return to natural rhythm, restoring sleep cycles that modern light exposure has badly disrupted. It's the perfect preparation for body and mind to receive whatever comes after it.
It is, and honestly it's less well understood scientifically too. What we do know is that it appears to act as a neuromodulator, and it's sometimes referred to as a visionary neurotransmitter, influencing consciousness and altered states of awareness. Research suggests it may have neuroprotective properties and could play a role in meditation or dream states, and some studies indicate it may affect dopamine regulation and serotonin transport. Experientially, this is where the brain seems to begin opening doorways that stay closed under constant light - pinoline appears to bridge ordinary consciousness and the more extraordinary states that follow.
That's right, it's gained a lot of attention through the growing popularity of Bufo alvarius - the Sonoran desert toad - though its function in the body still isn't confirmed by modern science. What's notable is that both forms of DMT are released in large amounts at birth and at death. Rick Strassman's book DMT: The Spirit Molecule explores some of the hypotheses around what these compounds might actually be for. In terms of effects, people report altered states of consciousness, mystical experiences, and shifts in perception and sense of self. This is often where the journey becomes particularly profound, because the brain is producing the same compounds it produces at life's most significant thresholds.
This one remains genuinely debated in the literature. We know it's naturally produced in small amounts in the human brain, and in many plants too. At this depth people sometimes report deeper levels of visual experience, and occasionally the externalisation of internal energies gets perceived as something like encountering entities. I always want to be careful here though - this is nowhere near the intensity of smoking DMT. It's much more gradual and subtle, and there's huge variation between individuals; some people have minimal or no visions at all. Generally, longer stays do increase the likelihood of disorientation and visual phenomena, which is exactly why we take the screening and pacing of longer retreats so seriously. Both forms of DMT are currently being studied for potential therapeutic use in depression, PTSD, and addiction, for what it's worth.
Serotonin is stimulated by light and speeds brain activity up into the higher alpha and beta wave frequencies - it's what keeps you analytical, active, oriented to the day. Melatonin does the opposite, slowing brain activity into deeper states. That creates a fundamental shift in consciousness: light-driven serotonin keeps you in active, analytical mode, while Darkness-produced melatonin opens access to deeper, more receptive layers of awareness - the layers that seem to matter for healing, creativity, and spiritual insight. In our light-saturated world we've become almost entirely dependent on serotonin's activation. Darkness offers something we've largely lost access to - the depth that melatonin provides.
The single biggest driver is artificial light after dark. The pineal gland evolved to receive one simple signal - Darkness means night - and we've comprehensively broken that signal. There's research comparing red and blue LED exposure that found after two hours, blue light kept melatonin suppressed at around 7.5 picograms per millilitre, while red light allowed natural recovery up to 26 - a difference that's especially pronounced in younger people. Every screen we look at in the evening floods the retina with exactly the wavelength, in the 450 to 500 nanometre range, that's most effective at suppressing nocturnal melatonin, delaying sleep onset, fragmenting sleep, and reducing deep slow-wave sleep. Diet compounds it - melatonin is synthesised from dietary tryptophan, and disrupted light-dark cycles, night shift work, and being overweight are all consistently linked to lower levels. Magnesium and B6, which are key co-factors in that synthesis pathway, are widely deficient in modern diets too, and alcohol and chronic stress suppress production further. On top of all that, endogenous melatonin naturally declines with age as retinal light perception and pineal activity diminish - so this decline is now landing on top of an already-suppressed baseline for most people.
Exactly. Rather than restoring the body's own production, we've built a mass supplementation industry that bypasses it instead. What the research ultimately confirms is something anyone who's spent time in a properly Dark environment already knows in their bones. Modern humans are living in a state of chronically suppressed melatonin. When complete Darkness is finally restored - when the pineal gland receives an unambiguous, uninterrupted signal, often for the first time in years - the resulting flood of endogenous melatonin isn't some bonus extra. It's a return to a biological baseline we've collectively lost.
The hypnagogic state is that drowsy, drifting transition from wakefulness into sleep. The hypnopompic state is the reverse - coming back from sleep into waking. During both, brain waves shift from the faster beta waves of normal waking consciousness, through alpha, toward slower theta waves - and theta is the range associated with deep meditation, creativity, and altered states. The brain is essentially caught between frequencies, which creates a genuinely unique neurological window. What Darkness does is extend the time spent in these states, which may well explain why Darkness practices show up in spiritual traditions worldwide. In there people often report vivid, sometimes surreal or symbolic imagery, a dreamy state between conscious and unconscious mind, spontaneous insights or creative breakthroughs, and unusual auditory or tactile experiences. The usual filters of rational consciousness relax, and that can open access to deeper layers of awareness and insight.
It connects directly. In Darkness, you may not just be having experiences - you may be participating in the reshaping of the very organ that creates all your experiences. Extended Darkness may enhance neuroplasticity, the brain's ability to form new connections and restructure existing ones. Research suggests that significant environmental changes, of which entering complete Darkness is about as significant as it gets, can stimulate neuroplastic responses. What that means practically is that insights and transformations occurring in Darkness may become integrated into new neural pathways that persist after someone returns to light. I'll say plainly that more research is needed to fully understand the mechanisms here - this is an area where the science is still catching up to the experience.
What we think happens is that Darkness heightens the role of the hypothalamus, which governs circadian rhythms throughout the body. It doesn't just regulate day-night hormones - it orchestrates many of the body's other internal clocks too. The hypothalamus directs function through two main pathways, one of which runs via the brainstem into the Autonomic Nervous System - the ANS - which itself splits into the sympathetic branch, your fight-or-flight response, and the parasympathetic branch, your rest-and-digest function. Melatonin from the pineal gland quietens the sympathetic nervous system and allows for the daily rejuvenation of mind and body. By what's sometimes called the 'pinoline stage', around days three to five, the nervous system starts to become aware of itself. Around days six to eight, as the pineal gland begins producing 5-MeO-DMT, it's said to switch on as much as 40% more of the cerebral cortex, deepening that self-awareness further. This is where the nervous system's bio-computer, so to speak, can start to reprogramme itself - potentially giving rise to profound meditative trance states and even, in some reports, telepathy within the emotional and intuitive bodies. The 5-MeO-DMT then prepares the nervous system for the pineal gland's DMT production around days nine to twelve, in psychoactive milligram doses.
